چکیده

Question IL-18 is a pro-inflammatory cytokine produced by a variety of myeloid and non-hematopoietic cells. It is canonically associated with enhancing interferon gamma (IFNg) and cytotoxicity in collaboration with IL-12p70, IL-15, or type I IFN. However, in other contexts IL-18 can promote IL-17, IL-22, or allergic responses. Macrophage Activation Syndrome (MAS) is a sepsis-like syndrome that has been associated with elevated serum IL-18 in systemic Juvenile Idiopathic Arthritis, Stills disease, and XIAP-deficiency. We sought to characterize IL-18 and associated cytokines in a cohort of patients with a variety of monogenic or complex autoinflammatory syndromes.

Whole exome sequencing in systemic juvenile idiopathic arthritis

Introduction Systemic juvenile idiopathic arthritis (sJIA) shares clinical features with classic monogenic autoinflammatory diseases, characterised by fevers, arthritis and evanescent rashes. Disease exacerbations are associated with elevated serum cytokine levels including 1L-1b, IL-6, and IL-18; and clinical response to anakinra, canakinumab and tocilizumab suggests that cytokine dysregulatio...

MRP8 and MRP14, phagocyte-specific danger signals, are sensitive biomarkers of disease activity in cryopyrin-associated periodic syndromes.

OBJECTIVES To assess the sensitivity of the phagocyte-specific molecules myeloid-related protein (MRP) 8 and MRP14 (calprotectin) for monitoring disease activity during anti-interleukin (IL)-1 therapies in patients with cryopyrin-associated periodic syndromes (CAPS), including familial cold autoinflammatory syndrome (FCAS), Muckle-Wells syndrome (MWS) and chronic infantile neurological, cutaneo...

Free circulating interleukin-18 is increased in Schnitzler syndrome: a new autoinflammatory disease?

Schnitzler syndrome is a rare disease characterised by chronic urticaria and arthralgia. The recent evidence that the IL-1 receptor antagonist IL-1Ra could induce rapid and complete remission of Schnitzler symptoms has pointed to IL-1 as a major pathological factor in this disease. To examine the possibility that Schnitzler syndrome may be considered to be an autoinflammatory disease, in this s...

The hyperferritinemic syndromes and CD163: a marker of macrophage activation.

F erritin is a key protein in iron metabolism. Its involvement in iron detoxification and iron storage is known, but recently, a new role in the pathogenesis of different autoimmune and autoinflammatory syndromes was hypothesized [1]. Four immune mediated conditions sharing the feature of marked hyperferritinemia have been gathered under the single term “Hyperferritinemic Syndromes” [1]. Such s...

Chronic, Systemic Interleukin-18 Does Not Promote Macular Degeneration or Choroidal Neovascularization

Age-related macular degeneration (AMD) and its complications remain major causes of ocular disability. We have followed with interest the debate regarding whether IL-18 contributes to, or protects from, AMD and subsequent choroidal neovascularization (CNV). Two articles published in 2012 approached the role of this inflammasome-activated cytokine with conflicting conclusions. Tarallo et al. obs...
